L Tryptophan is the precursor to Serotonin, a neurotransmitter in the brain, which is deficient in depression and naturally enhances sleep. Each capsule of Pure L-Tryptophan has 500 mg. of the highest quality USP Pharmaceutical Pure Grade L - Tryptophan available for professional use.
Directions: As a sleep aid, Take 1-3 capsules 30 minute before bedtime, during the night if required, or as needed. Tryptophan And Sleep
Sleep Deprivation is a serious concern for many, and low Serotonin levels are often the cause of this problem. Tryptophan converts to Serotonin helping to induce relaxation and naturally enhancing sleep. Two capsules 30 minutes prior to bedtime are recommended. If necessary, Tryptophan can be taken during the night. In extreme problems, Tryptophan should be taken early evening and again at bedtime. Vitamin B-6, along with increasing dietary complex carbohydrates are important for proper metabolism and increasing effectiveness.

Mood Enhancement
It has been well documented that low Serotonin levels and low Tryptophan levels have been associated with depression. Tryptophan naturally converts to this neurotransmitter. If depression also involves Catecholamine biochemistry, then taking Tryptophan in a regimen along with Tyrosine is recommended. Since they compete for uptake in the brain, and because Tryptophan induces sleep, it is recommended that Tryptophan be taken prior to dinner and again prior to bedtime. Other Aspects of L-Tryptophan and Serotonin Autism
Low levels of Tryptophan have been documented in autistic children, indicating that there may be a defect in the Tryptophan-Serotonin metabolism in the brain.
Pain Tolerance
Increased Serotonin levels have an effect on increasing pain tolerance, especially for chronic pain caused from Arthritis and Lower Back Problems. In one study, Tryptophan was administered at the dose of 1-500 mg. capsule every 2 1/2 hours(total 6 per day) resulting in a 50% reduction in pain intensity. A high carbohydrate diet is recommended to increase Tryptophan metabolism, along with Vitamin B-6. D-Phenylalanine is also recommended in some cases to enhance results and supplementation with Montiff D Phenyl Relief (D Phenylalanine) is suggested.
Appetite Control
Some studies suggest that administering Tryptophan helps control appetite, for those who are on a weight reduction program.
Chemical Addiction
Cocaine addiction causes a deficiency of L-Dopamine and causes both Cathecholamine and Serotonin neurotransmitters imbalances. WHAT IS TYROSINE ?

Tyrosine is an amino acid, which is made in the body from Phenylalanine.

WHAT IS D-PHENYLALANINE?
D-Phenylalanine is the Dextrorotatory form of the amino acid, Phenylalanine. The L-forms (Levorotatory) of amino acids are ones that are bioavailable and are for protein synthesis and regulatory functions. They are like mirror images of one another, however, D-Phenylalanine is not absorbed into the body, but acts as an inhibiting agent to the enzymes that are responsible for the breakdown of endorphins, which control pain perception.

WHAT ARE ENDORPHINS?
Endorphins (and enkephalins) are naturally occurring opiate-like peptides, which control pain perception and produce an analgesic effect. Their effects are only of a short duration because of metabolic enzymes, especially Carboxypeptidase A, which inhibit increased endorphin Production. D-Phenylalanine prohibits these enzymes, allowing increased endorphin Production, and prolonged and intensified analgesic affects.

HOW DOES D-PHENYLALANINE EFFECT PAIN?
Low levels of endorphins have been noted in those suffering from chronic pain, and supplementation with D-Phenlyalanine helped to increase these levels. By prohibiting Carboxypeptidase A from preventing endorphin Production, the body can continue to increase its own natural pain tolerance mechanism. In studies with chronic pain patients, the pain threshold has been enhanced and prolonged. This has been effected with various chronic pain conditions, including back pain and Osteoarthritis.

D-PHENYLALANINE AND ACUPUNCTURE
Pre-administration of D-Phenylalanine enhances the effects of acupuncture on back and other pain. Up to 2 grams 1 hour prior to acupuncture treatment may result in more pain relief and for longer periods of time.

DENTAL PAIN
Two grams of D-Phenylalanine has been given 1 hour prior to dental extractions, increasing the pain threshold and for a prolonged period of time.

WHY NOT DLPA (A COMBINATION OF BOTH D-PHENYLALANINE AND L-PHENYLALANINE)? D- PHENYLALANINE is the form of Phenylalanine that effects endorphin Production. L-Phenylalanine has other functions in the body. Concentrations of the pure D-Phenylalanine will assist in pain tolerance to those needing this function. By combining the two, the concentration of the effective form of D-Phenylalanine would be less, and therefore more would be required to achieve results. WHAT IS CITRULLINE ?

Citrulline, a non-essential amino acid synthesized in the intestinal tract from Glutamine, converts to Arginine in a biochemical process involving Aspartic Acid and two necessary enzymes, Ornithine Transcarbamoylase and Argininosuccinate Synthetase. As a precursor to Arginine, It has a sparing effect on Arginine enabling an increase in Nitric Oxide production. Nitric Oxide (NO) is very important for endothelial support and cardiovascular health. Citrulline increases energy and stimulates the immune system, as well.

Citrulline is also an essential component of the Urea Cycle, and functions along with Ornithine and Arginine to convert ammonia into Urea. High ammonia levels are extremely toxic and can cause liver damage and neurological impairment. Supplementation with Citrulline has been very beneficial in a genetic disorder called Ornithine Transcarbamoylase (OTC) Deficiency.

CITRULLINE AND CARDIOVASCULAR HEALTH

Citrulline is a precursor to Arginine, which has been shown to increase Nitric Oxide. Since up to 80% of Arginine utilized in the liver and kidneys, is only 20% available for cardiovascular support. Supplementing with Citrulline produces a sparing effect allowing for additional Arginine production intracellularly for cardiovascular function. Nitric Oxide (NO), the endothelial relaxing factor (EDRF), is a small molecule made from Arginine. It plays an essential role in the regulation of vascular homeostasis, modulates cardiac contractility, regulates vascular tone, and helps in preventing vessel injury and Atherosclerosis. Oxidative stress (free radical damage) creates injury to endothelial cells. Citrulline converts to Arginine in the vascular cells, and thus increases Nitric Oxide production, which can help to restore endothelial cell function. The role of Nitric Oxide and Arginine supplementation, as well as the role of Citrulline in endothelial function has been well-documented in medical studies. Dr. S. Gross (Cornell U. Med. College) in his publication in the J. Biol. Chem., 1997 suggest the ratio of 3 Arginine to 1 Citrulline for Nitric Oxide production.

***Those with active Herpes or similar viruses who need cardiovascular support should reverse the Citrulline/Arginine ratio to 3 Citrulline to 1 Arginine, since Arginine can stimulate the Herpes. This ratio can help increase Arginine intracellularly. Lysine supplementation may also be advisable in some cases.

UREA CYCLE FUNCTION

The Urea Cycle functions to remove toxic ammonia by converting it to Urea (primarily in the liver). Ammonia is a highly toxic substance synthesized from protein and amino acid catabolism, and is also formed in the intestinal tract. It is also highly elevated in 6 Urea cycle disorder of enzyme deficiencies (OTC, CPS-Carbamoyl Deficiency, ALD-Arginosuccinic Aciduria, AG-Arginase, ASD-Citrullinemia and NAGS-N-Acetylglutamate Synthestase) which are genetic biochemical abnormalities that can be fatal. Athletes also tend to have somewhat elevated ammonia levels.

The Urea cycle functions in a complicated biochemical process in which Ornithine, Citrulline, Aspartic Acid, Argininosuccinate Acid, Fumeric Acid, and Arginine, along with the necessary enzymes and Carbon Dioxide convert ammonia to Urea. All of these are components and function as the Urea cycle. Since high levels of ammonia are extremely toxic and may lead to death, it is essential for the Urea cycle to perform properly. OTC is a genetic disorder of the Urea cycle in which there is a Citrulline deficiency.

ORNITHINE TRANSCARBAMOYLASE (OTC) DEFICIENCY

This is a serious genetic Urea cycle disorder effecting an estimated 1 in 20,000 births. Ornithine Transcarbamoylase is the second enzyme in the Urea cycle and is necessary to produce Citrulline. A deficiency of this enzyme causes a lack of Citrulline, thus the Urea cycle can't function properly and the result is hyperammonemia. It has been linked to SIDS deaths in newborns, and Reye's syndrome in children, since it is often undiagnosed. Symptoms of the neonatal onset disorder may appear in the first 24 hours of life with vomiting and lethargy, followed by neonatal seizures and coma. This disorder often presents in males in the neonatal period, although partial enzyme deficiencies also affect both males and females in childhood and adulthood. This late onset form of the disorder may present in childhood with recurrent symptoms including lethargy, migraine-like headaches, persistent vomiting, dehydration, hyperactivity, bouts of screaming, acute taxia, retardation and coma. There are cases of female carriers of this disorder developing symptoms as adults as well. Left untreated, it is 100% fatal, and there is no know cure. Limiting dietary protein, special formulas and Citrulline supplementation are advised to increase Urea cycle function and reduce ammonia.

WHAT IS TRYPTOPHAN?

PYRIDOXAL- 5'-PHOSPHATE
Pyridoxal- 5'-Phophate (the enzymatic - active form of Vitamin B-6) is essential for Tryptophan metabolism and conversion to Serotonin. Most B-6 supplements are Pyridoxine, which must be converted in the liver to Pyridoxal-5'-Phosphate for it to be properly utilized. RIBOFLAVIN-S'-PHOSPHATE

It helps convert Pyridoxine to the active Pyridoxal- 5'-Phosphate. A deficiency of Riboflavin may cause deficiency of P-5'-P, resulting in neurotransmitter imbalances and deficiencies, including preventing the conversion of Tryptophan to Serotonin. Inactive Riboflavin (B-2) has to be converted in the liver to Flavin Mononucleotide (FMN) & Flavin Adenine Dinucleotide (FAD), and Flavokinase production is also necessary in order for B-2 to be activated and properly utilized. Sufficient amounts do not convert in many individuals, and a deficiency of active Riboflavin can cause significant behavioral problems including psychopathy, depression, hysteria, and hypomania. In addition, Psychopharmalogical drugs, including chlorpromazine, imipramine, and amiltriptyline actually inhibit the formation of FMN & FAD. Riboflavin-5'-Phosphate is the active form of Riboflavin and it is included in Tryptophan-Plus or increased effectiveness.

TRYPTOPHAN-PLUS AND SLEEP
Sleep deprivation is a serious concern for many, and low Serotonin levels are often the cause of this problem. Enteric Preparation

PYRIDOXAL-5-PHOSPHATE (P-5’-P-Active, Co-enzyme B-6) is the active, co-enzymatic form of B-6. Many individuals do not convert sufficient amounts of this important B vitamin for proper metabolism and functions, which can result in a B-6 deficiency and many health problems. Pyridoxal-5'-Phosphate is necessary for amino acid absorption and metabolism and is involved in the synthesis and catabolism of all protein. It is especially important in the metabolism of Tryptophan, Tyrosine, Glycine, Glutamic Acid, Cysteine, Cystine, and Methionine. Insufficient P-5’-P may cause abnormal metabolites such as Homocysteine (which is associated with heart attacks and strokes). Pyridoxal- 5'-Phosphate is utilized by over 60 enzymes, and it is also essential for fat and carbohydrate metabolism. Three Phos-B contains 20 mg. of Pyridoxal-5'-Phosphate.

DEFICIENCIES AND SYMPTOMS
• Numbness and tingling of limbs, depression, nervousness, cracks around mouth and eyes, water retention, fatigue. Headache, nausea, acne, and inflammation of the gums are also symptoms.
• Low levels of P-5’-P have also been noted with low plasma levels of Tyrosine and Tryptophan, which correlate to deficiencies in serotonin and catecholamine neurotransmitters. These are associated with depression, anxiety, stress and sleep deprivation.
• Cigarette smoking, alcohol and many drugs, such as antibiotics and birth control pills, can cause a B-6 deficiency.
• Carpal tunnel syndrome is also associated with a B-6 deficiency.
• P-5-P deficiencies have been noted in those whose seizures, which are associated with hyperprolinemia type II. The research reported that these seizures may be preventable with long term supplementation.
• Elderly people tend to be deficient in P-5’-P.
• Pregnant women are generally deficient and supplementation may decrease morning sickness.
• Deficiency may cause in decrease of mineral absorption and utilization, including Calcium.

RIBOFLAVIN-5-PHOSPHATE (Co-enzyme form of B-2) is the active form of B-2, which is essential for ATP production and energy metabolism, maintenance of the nervous system, carbohydrate metabolism, and is involved in Glutathione synthesis. Riboflavin is also important maintaining healthy eye tissue, skin, mucous membranes and fetal growth and development. It also converts B-6 and folic acid into their coenzymatic forms. Riboflavin is found in milk, eggs, lean meat and broccoli, as well as and enriched breads and cereals. Since Riboflavin is destroyed by light, milk purchased in plastic jugs have decreased amounts of Riboflavin. Three Phos-B enteric preparation contains 3 mg. of Riboflavin- 5'Phosphate per tablet.

DEFICIENCIES AND SYMPTOMS
• Fissures around mouth, nose and ears, as well as eye irritation and photophobia.
• Older people tend to have Riboflavin deficiencies.
• Deficiencies in Riboflavin cause Glutathione deficiencies, leading to increased incidence of cataracts.
• Since Riboflavin helps convert Pyridoxine to the active Pyridoxal 5'Phosphate, a deficiency of Riboflavin may cause a deficiency of P-5’-P. This could result in amino acid and neurotransmitter imbalances and deficiencies.

THIAMINE-5-PHOSPHATE (Co-enzyme form of B-1) Thiamine is essential for almost every cellular reaction in the body and is necessary for proper development and growth. It is important for healthy skin and hair, as well as blood production, immune function, and metabolism of alcohol. In addition it is especially important for cardiovascular health, proper nerve function and the synthesis of the neurotransmitter, acetylcholine, which is involved in memory and other brain function. Cognitive dysfunction is often associated with a deficiency, as well as lack of appetite. Many older people are deficient in this B vitamin, as well. It is not stored in great amounts in the body and must be obtained daily from the diet or supplementation. Some of the food sources of Thiamine are: pork, organ meats, brewer's yeast, oatmeal, grains, poultry, nuts, avocado, thiamine enriched flour and cereal, and some vegetables such as spinach and cauliflower. Heat easily destroys Thiamine and a considerable amount is lost in the cooking process of the foods, including in the water of cooked vegetables. Sulfate food destroys 131 and alcohol reduces the absorption of Thiamine. Each Three Phos-B contains 2 mg. of the co-enzymatic form of Thiamine.

DEFICIENCIES AND SYMPTOMS
• Beriberi -Dry Beriberi (symptomatic of severe muscle wasting) and Wet Beriberi (in which edema is present).
• Brain and nervous systems are effected. Some symptoms are poor memory and confusion, irritability, depression and inability to concentrate with a reduced attention span.
• Tingling and numbing of the extremities, as well as painful calf muscles.
• Cardiovascular -The heart requires Thiamine and a deficiency may exhibit in symptoms such as cardiac failure, ankle swelling, fatigue and heart enlargement.
• Alcohol Related. Since alcohol reduces absorption, alcoholics and binge drinkers are prone to thiamine deficiency, resulting in mental confusion and lack of balance in walking, referred to as a staggering gate. • Athletes with strenuous activities require additional Thiamine, and when supplemented have less fatigue and more energy. Physical activity also increases the requirements for B-2 and B-6.

WHAT IS ARGININE?
Arginine is synthesized in the intestinal tract from Citrulline, and its major food sources are nuts, (peanuts, cashews, almonds, walnuts and pecans) although it is also abundant in chocolate, carob, and edible seeds. It is the precursor for Ornithine and is a vital part of the urea cycle, being involved in the detoxification and elimination of ammonia. It is also important for Nitric Oxide production, (necessary for endothelial health and vasodilation), protein synthesis, wound healing, immune function and has a secretalogical effect on hormones. Eighty percent of seminal fluid is made of Arginine, and it plays a role in the reproductive system of females as well.
